Mizzou. We've been Tigers longer than both Auburn and LSU.
Founding year:
Mizzou--1839
Auburn--1856
LSU--1860
Clemson--1889
Memphis--1912
Adoption of "Tigers" nickname/first football season:
Mizzou--1890
Auburn--1892
LSU--1893
Clemson--1896
Memphis--1914
Mizzou was the "Tigers" before LSU and Auburn anyway you look at it.
Edited to add Memphis and Clemson for all the people throwing them out there.

This game was for the right to be called the "real Tigers" and advertised as such. It was settled back in 1978 at the Liberty Bowl. Mizzou beat LSU 20-15 on that beautiful day. So, until LSU takes it...it belongs to Mizzou. Done deal, end of discussion.
Auburn is confused between birds and mammals. Bless their heart!

The guys who have a live tiger on campus, not the team with the split personality....war eagle one game, tigers the next. I think of auburn as the war eagles...their tiger mascot is more for fun.

The University of Memphis has a live tiger that actually attends their home games.

And Princeton was the Tigers before all of us....

This is true, although they never formally adopted the nickname as the other universities did. Earliest references to Princeton Tigers were in the 1880's but wasn't really set in stone until the mid 1910's. LINK
Yes I'm being nitpicky, so I'll still give Princeton credit for being first.

You think that's bad, look at Notre Dame.
"Notre Dame" is French. The university is in the USA, and their mascot is the fighting Irish.
Goodness, hahah.
