For hotels, if you are flying out of St. Louis then Kingdom City or Fulton are close to I-70 but your options are limited. Kingdom City is just basically a truck-stop town, Fulton is a little bigger with a few restaurant options.
Jefferson City is bigger but a little more out of the way unless you are flying out of Columbia, in which case that is where I would stay. There is a Doubletree close to downtown which is decent and within walking distance to some restaurants and bars.

This is all good info from my fellow Mizzou posters, but I agree with the Hog. I wouldn't advise staying in Boonville, Moberly or Fulton unless you get pretty desperate.
Jeff City isn't a terrible option and is an easy 20-25 minute trip to Columbia.
I would also say fly into STL instead of KC if possible. KC is more of a hassle and a longer trip to Columbia.

Boonville is a great town. Frederick Hotel has neat old hotel rooms and Glenn's restaurant, a famous cajun place formerly of Columbia, is in the hotel and serves up some fine food. Never been in any of the hotel rooms at the casino.
